33. What are your gene delivery vehicles, the target cells, and how you would apply the target cells.

Answers:

A. Age-related Macular Degeneration (AMD):We plan to inject clinical-grade AAVs encoding the reprogramming factors directly into the eye. The specific AAV serotype has already been calibrated in the lab. We’re collaborating with ophthalmologists at Hadassah hospital who routinely perform this injection technique.

B. Skin:We will take a biopsy from behind the patient’s ear, expand the cells in the lab, and deliver the factors using modified RNAs to control their expression. Our long-term goal is to develop a topical lotion that can deliver the factors directly to the dermis.

C. Immune System: We intend to isolate CD34+ cells from peripheral blood and introduce the factors using clinical-grade AAVs. The serotype for this has also been calibrated in the lab.

34. Where is your technology coming from?

Answer: Ten Years of research: Since 2014, the Hebrew University of Jerusalem has studied and developed a unique patented set of factors/genes that can rejuvenate cells, which are believed to be superior to those of our competitors. Prof. Yosef Buganim, a world leader in the field of stem cells and reprogramming, was the first to discover a combination of genes that can convert adult cells into placental stem cells. Given that the chronological age of the placenta is zero (embryonic stage), Prof. Buganim’s method can also rejuvenate cells through site-specific alteration of epigenetic marks.

His research demonstrated that this combination of factors is particularly potent in targeting the genome of adult cells, achieving up to 50% age reversal in adult skin cells while preserving their identity. This age reversal was associated with improved function and healthier cells. Moreover, unlike Yamanaka factors, which carry a tumorigenic risk (formation of teratoma, a benign tumor), Prof. Buganim’s combination is safer.
